Summary : Oxidative Stress and Dietary Antioxidants in Neurological Diseases provides an overview of oxidative stress in neurological diseases and associated conditions, including behavioral aspects and the potentially therapeutic usage of natural antioxidants in the diet. The processes within the science of oxidative stress are described in concert with other processes, such as apoptosis, cell signaling, and receptor mediated responses. This approach recognizes that diseases are often multifactorial and oxidative stress is a single component of this. The book examines basic processes of oxidative stress—from molecular biology to whole organs—relative to cellular defense systems, and across a range of neurological diseases. Sections discuss antioxidants in foods, including plants and components of the diet, examining the underlying mechanisms associated with therapeutic potential and clinical applications. Although some of this material is exploratory or preclinical, it can provide the framework for further in-depth analysis or studies via well-designed clinical trials or the analysis of pathways, mechanisms, and components in order to devise new therapeutic strategies. Very often oxidative stress is a feature of neurological disease and associated conditions which either centers on or around molecular and cellular processes. Oxidative stress can also arise due to nutritional imbalance during a spectrum of timeframes before the onset of disease or during its development. Offers an overview of oxidative stress from molecular biology to whole organs Discusses the potentially therapeutic usage of natural antioxidants in the patient diet Provides the framework for further in-depth analysis or studies of potential treatments

Summary : The brain is the most complex organ in our body. Indeed, it is perhaps the most complex structure we have ever encountered in nature. Both structurally and functionally, there are many peculiarities that differentiate the brain from all other organs. The brain is our connection to the world around us and by governing nervous system and higher function, any disturbance induces severe neurological and psychiatric disorders that can have a devastating effect on quality of life. Our understanding of the physiology and biochemistry of the brain has improved dramatically in the last two decades. In particular, the critical role of cations, including magnesium, has become evident, even if incompletely understood at a mechanistic level. The exact role and regulation of magnesium, in particular, remains elusive, largely because intracellular levels are so difficult to routinely quantify. Nonetheless, the importance of magnesium to normal central nervous system activity is self-evident given the complicated homeostatic mechanisms that maintain the concentration of this cation within strict limits essential for normal physiology and metabolism. There is also considerable accumulating evidence to suggest alterations to some brain functions in both normal and pathological conditions may be linked to alterations in local magnesium concentration. This book, containing chapters written by some of the foremost experts in the field of magnesium research, brings together the latest in experimental and clinical magnesium research as it relates to the central nervous system. It offers a complete and updated view of magnesiums involvement in central nervous system function and in so doing, brings together two main pillars of contemporary neuroscience research, namely providing an explanation for the molecular mechanisms involved in brain function, and emphasizing the connections between the molecular changes and behavior. It is the untiring efforts of those magnesium researchers who have dedicated their lives to unraveling the mysteries of magnesiums role in biological systems that has inspired the collation of this volume of work.

Summary : The plant-derived polyphenol curcumin has been used in promoting health and combating disease for thousands of years. Its therapeutic effects have been successfully utilized in Ayurvedic and Traditional Chinese Medicine in order to treat inflammatory diseases. Current results from modern biomolecular research reveal the modulatory effects of curcumin on a variety of signal transduction pathways associated with inflammation and cancer. In this context, curcumin’s antioxidant, anti-inflammatory, anti-tumorigenic, and even anti-metastatic activities are discussed. On the cellular level, the reduced activity of several transcription factors (such as NFkB or AP-1) and the suppression of inflammatory cytokines, matrix degrading enzymes, metastasis related genes and even microRNAs are reported. On functional levels, these molecular effects translate into reduced proliferative, invasive, and metastatic capacity, as well as induced tumor cell apoptosis. All these effects have been observed not only in vitro but also in animal models. In combination with anti-neoplastic drugs like Taxol, kinase inhibitors, and radiation therapy, curcumin potentiates the drugs’ therapeutic power and can protect against undesired side effects. Natural plant-derived compounds like curcumin have one significant advantage: They do not usually cause side effects. This feature qualifies curcumin for primary prevention in healthy persons with a predisposition to cancer, arteriosclerosis, or chronic inflammatory diseases. Nonetheless, curcumin is considered safe, although potential toxic effects stemming from high dosages, long-term intake, and pharmacological interactions with other compounds have yet to be assessed. This Special Issue examines in detail and updates current research on the molecular targets, protective effects, and modes of action of natural plant-derived compounds and their roles in the prevention and treatment of human diseases.

Summary : Inflammation has invaded the field of psychiatry. The finding that cytokines are elevated in various affective and psychotic disorders brings to the forefront the necessity of identifying the precise research domain criteria (RDoCs) that inflammation is responsible for. This task is certainly the most advanced in major depressive disorders. The reason is that a dearth of clinical and preclinical studies has demonstrated that inflammation can cause symptoms of depression and conversely, cytokine antagonists can attenuate symptoms of depression in medical and psychiatric patients with chronic low grade inflammation. Important knowledge has been gained on the symptom dimensions that inflammation is driving and the mechanisms of action of cytokines in the brain, providing new targets for drug research and development. The aim of the book “Inflammation-Associated Depression” is to present this field of research and its implications in a didactic and comprehensive manner to basic and clinical scientists, psychiatrists, physicians, and students at the graduate level.

Summary : "Mechanisms of Neuroinflammation" book explains how the neuronal cells become swollen at the moment of the blood-brain barrier disruption and how they lose their immunological isolation. A cascade of cytokines and immune cells from the bloodstream enters the nervous system, inflaming neurons and activating the glia. This produces a neuroinflammatory process that can generate different neurodegenerative diseases. Better understanding of mechanisms that are activated at the time when the damage to the brain occurs could lead to the development of suitable therapies that revert the neuronal inflammation and thus prevent further damage to the nervous system.

Summary : Role of the Mediterranean Diet in the Brain and Neurodegenerative Disease provides a comprehensive overview of the effects of all components of the Mediterranean diet on the brain, along with its beneficial effects in neurodegenerative diseases. It covers topics on neurodegenerative diseases (Alzheimer disease (AD), Parkinson disease, (PD) Huntington disease (HD) and Amyotrophic Lateral Sclerosis (ALS), also providing information on how cardiovascular disease, Type 2 Diabetes, and Metabolic Syndrome become risk factors for neurodegenerative diseases. This book focuses on how the Mediterranean diet suppresses oxidative stress and neuroinflammation in neurodegenerative diseases as well as signal transduction. The Mediterranean diet is characterized by the abundant consumption of olive oil, high consumption of plant foods (fruits, vegetables, pulses, cereals, nuts and seeds); frequent and moderate intake of wine (mainly with meals); moderate consumption of fish, seafood, yogurt, cheese, poultry and eggs; and low consumption of red meat and processed meat products. High consumption of dietary fiber, low glycemic index and glycemic load, anti-inflammatory effects, and antioxidant compounds may act together to produce favorable effects on health status. Collective evidence suggests that Mediterranean diet not only increases longevity by lowering cardiovascular disease, inhibiting cancer growth, but also by protecting the body from age-dependent cognitive decline.
