Summary : Major advancements in the field of in situ molecular pathology have occurred since publication of the first edition. In Situ Molecular Pathology and Co-expression Analyses, Second Edition, continues to teach both the molecular basis for the improvements and the actual protocols. This is the unique feature that separates it from the pack of other "cook-book" type approaches. The fields of in situ hybridization and immunohistochemistry have expanded rapidly where computer-based analyses systems have greatly expanded the power of these methods. Further, knowledge of the marked improvements in the reagents themselves since the first edition can make the difference of excellent versus misleading data. The automated platforms require that researchers and diagnostic biomedical investigators have a good understanding of the basics of in situ based tests, protocols, and biochemistry for troubleshooting in order to maximize the use of these platforms. This second edition focuses attention on straightforward protocols used to simultaneously detect two or more proteins/nucleic acids within intact tissue by doing co-expression analyses. Practicing molecular pathologists, diagnostic pathologists, laboratory directors, and toxicologists, as well as clinicians and researchers in training, will benefit from this clear presentation of protocols and theoretical framework. Data derived from in situ hybridization and immunohistochemistry. Explains the theory and foundation of immunohistochemistry and in situ hybridization and presents easy-to-follow experimental protocols with tricks of the trade Includes two new chapters: Recent improvements in immunohistochemistry and in situ hybridization, Quality control for immunohistochemistry and in situ hybridization: How to know if the color change is signal or background The second edition also includes a detailed test to help one learn the basics of histologic interpretation of tissues and a separate detailed test in how to differentiate signal from background Includes chapter-ending summaries of Key Points to Remember, bringing beginners up to speed with any seasoned veteran in the field Thoughtfully tackles the molecular basis if IHC and ISH, along with application of that knowledge to improving the techniques is significant

Summary : There has been an explosion of knowledge and enormous progress in the fundamental understanding of the biology of cancer in recent years. This has included the realisation that cancer occurs when normal cellular functions are disturbed leading to a malignant phenotype. Much research has focused on understanding the types of disturbances that can occur, the contribution that these abnormalities can make to the development and behavior of particular cancers and more recently, the recognition that these cellular and genetic abnormalities can provide rational targets for new therapeutic approaches. Information about the biology of cancers that occur in children has increased in parallel with these more general advances and this book is intended to provide a focus for readers who wish to have an understanding of our current state of knowledge. An international group of editors and contributors provide guidelines on the molecular biology and pathology of paediatric oncology, aimed at clinicians and scientists working in the specialty who wish to understand current developments in molecular pathology as applied to their field. The book is broad ranging review focusing on the impact of molecular and cytogenetic techniques on our understanding of the aetiology, clinical behaviour, diagnosis and management of paediatric cancer. The first section outlines the laboratory handling of tissue samples, theory and methodology of cytogenetic and molecular techniques and discusses predisposition syndromes. The second section highlights the application of cytogenetic and molecular methods in diagnosis and treatment of the major paediatric cancers.

Summary : Twenty years have passed since I became a professor in the First Department of Pathology, Hiroshima University School of Medicine. It is my great pleas ure that Molecular Pathology o[ Gastroenterological Cancer-Application to Clinical Practice has been published by Springer-Verlag Tokyo to commemo rate the 20th anniversary of my professorship. Seeing the academic achievements of our department during these 20 years, I am confident that we could establish a department of oncology to research the pathogenesis of human cancer through systemic application of a variety of molecular techniques. We have demonstrated that the develop me nt and progression of esophageal, gastric, and colon cancer require mul tiple alterations affecting DNA mismatch repair genes, oncogenes, and tumorsuppressor genes, and that common and uncommon genetic changes exist for esophageal, gastric, and colorectal carcinomas. In addition to these genetic changes, the majority of gastrointestinal cancers express telomerase activity, with overexpression of telomerase RNA, indicating a powerful addi tional tool for early detection of gastrointestinal cancer. By transferring these basic observations to the clinic, we now are able to make accurate cancer diagnoses, thus determing the grade of malignancy and patient prognosis. We also can identify patients at high risk for developing cancer and create new therapeutic approaches. In fact, we have routinely implemented a new molecular diagnosis strategy at the Hiroshima City Medi cal Association Clinical Laboratory since August 1993.

Summary : The complex landscape of breast cancer requires distinct strategies for the management of various molecular subtypes of this disease. Rapid advances in the field of molecular biology have been bewildering for those involved in its study and management. “Molecular Pathology of Breast Cancer” aims to close this knowledge gap by discussing comprehensively the evolution, biological basis and clinical applications with a focus on the “what, when, and how” of the most significant molecular markers known to date. These markers are evaluated in the context of genomic, transcriptomic and proteomic profiles, which is integral to the practice of precision medicine. The application of next generation sequencing (NGS) has provided new insights in the regulation of genomic and transcriptomic structure and function. Alterations in DNA such as mutations and single nucleotide polymorphisms (SNPs) have been correlated with outcomes and provide for novel therapeutic approaches. These NGS analyses have also revealed the extensive contributions of epigenetic mechanisms such as histone modifications, non-coding RNA and alternative splicing. All of these changes together contribute to alterations in proteome. Newer assays that allow greater stability and analytical consistency are emerging. These alterations in tumor profiles can be also now detected by imaging techniques. The heterogeneity of both tumor and tumor microenvironment, an inevitable reality, is discussed in detail with particular focus on cancer stem cells and immune signaling. A chapter is dedicated to the emerging technology of “liquid biopsy”, which opens a novel approach for “continuous” monitoring of cancer that might be superior to conventional diagnostics, “Molecular Pathology of Breast Cancer” provides a quick and easy, not to mention essential, tour for clinicians, pathologists and scientists who are seeking to understand the integration of molecular biology into the diagnosis, prognosis and management of breast cancer.

Summary : The Maize Handbook represents the collective efforts of the maize research community to enumerate the key steps of standard procedures and to disseminate these protocols for the common good. Although the material in this volume is drawn from experience with maize, many of the procedures, protocols, and descriptions are applicable to other higher plants, particularly to other grasses. The power and resolution of experiments with maize depend on the wide range of specialized genetic techniques and marked stocks; these materials are available today as the culmination of nearly 100 years of genetic research. A major goal of this volume is to introduce this genetical legacy and to highlight current stock construction programs that will soon benefit our work, e. g. high-density RFLP maps, deletion stocks, etc. Both stock construction and maintenance are relatively straightforward in maize as a result of the ease of crossing and the longevity of stored seeds. Crossing is facilitated by the separate staminate (tassel) and pistillate (ear) flowers, a feature almost unique to maize. On the other hand, many of the genetic methodologies utilized with maize, including the precision of record keeping, can be adapted to other plants. Facile communication and a spirit of co-operation have characterized the maize genetics community since its earliest days. Starting in the 1930s, institutions such as annual Maize Genetics Cooperation Newsletter, the Maize Genetics Stock Center, and the annual maize genetics meeting provide continuity to the field.
